About Electron Microscopy, NeCEN, Leiden, The Netherlands

NeCEN is an open access facility for high resolution cryo-electron microscopy of biological samples. A state-of-the-art Titan Krios transmission electron microscope allows efficient and automated high resolution data collection in its two most broadly used forms, single particle analysis (SPA) and cryo-electron tomography (cryo-ET). These imaging techniques allow visualisation of biomolecular structures, such as proteins, macromolecular complexes, bacteria, and cell organelles at subnanometer resolution in close-to-native conditions. The facility is equipped with a BSL2 lab and microscope rooms.

Instruments Available:

Titan Krios 1 microscope has a X-FEG source and is equipped with a K3 bioquantum from Gatan. AFIS is possible, allowing SPA data collection speeds of up to 450 movies per hour. In a standard non AFIS data collection, speed varies from 60 (small holes, 1 shot per hole) to 180 (large holes, multiple shots per hole) movies per hour.

User Guide

The Titan Krios is a dedicated instrument for energy-filtered TEM (EFTEM) and cryo-electron tomography. This microscope is equipped with a K3 bioquantum DED. It is an instrument for both tomography and automated single particle acquisition.

Applications

NeCEN is open for applications from Dutch and international academia as well as industry. Access via the Instruct route is subsidized but not completely free. Contact us for details.
