The facility provides access to state-of-the-art cryo-EM instrumentation, including automated workflows for single particle data analysis, electron cryo-tomography, lamella preparation, and correlative imaging.

The cryo-EM facility offers the following workflows:

  • Sample preparation and Cryo-grid screening on the Arctica for samples prepared on Quantifoil grids (e.g. single particle analysis targets, exosomes, vesicles), including 2D classification for initial quality assessment.
  • High-end single particle data collection on the Titan Krios with cryoSPARC Live processing for pre-screened grids on the Titan Krios.
  • Tomogram acquisition on pre-screened samples such as large complexes, exosomes, and viruses.

Tailored support is provided for Instruct users, including project feasibility assessments, proposal preparation, and scheduling support.

Instruments Available:

This service provides cryo-electron tomography data acquisition of pre-screened plunge-frozen grids containing large complexes, viruses, or exosomes. It is intended for samples that do not require lamella preparation. Data are acquired on a Titan Krios microscope using automated tomography data acquisition workflows. 

Availability: Physical and remote (data collection under supervision) 

This service provides high-resolution single-particle cryo-EM data collection on pre-screened Quantifoil grids using automated acquisition workflows on a Titan Krios microscope. During the session, users can collect large datasets for structure determination, with real-time data processing available through cryoSPARC Live for immediate feedback on data quality and sample performance. This service is suitable for well-optimized samples that have already been screened and are ready for high-end data collection.

Average duration of visit: 2-3 days

This service supports users screening, and evaluating cryo-EM grids for high-resolution data collection. During the service session, up to 8 grids can be screened to assess overall sample suitability for high-end data acquisition, including check of ice quality, particle density and distribution. Optional grid preparation support can be provided upon request, depending on project needs and sample readiness. Initial data processing (motion correction, CTF estimation, and 2D classification) is included to assess data quality and guide next steps.

Used for sample screening (primarily purified protein complexes, vesicles), grid evaluation, EPU multigrid and cryoSPARC for fast evaluation. Tomography on thin samples is possible.
